Stability and method of maintaining microbial cultre from the Czech and foreign collections.

Cílem testu stability bylo ověřit a potvrdit navržený postup uchovávání baktérie C. necator a to fermentačním testem v poloprovozním měřítku. Jako zdroj sloužily bakterie z německé sbírky, které byly pomnoženy a uchovávány v lyofylizované formě. Procesní parametry jednotlivých kroků jako např. teplota, míchání, vzdušnění, pH, přetlak, feedovací (dávkovací) profily byly monitorovány a řízeny v průběhu celé fermentační části v rámci ověření navržení optimalizací a scale-upu parametrů v poloprovozním měřítku.

Task for this test was verification of the method of maintaining bacteria C.Necator in te pilot scale. As source were used bacteria from the collection which were cultivated and maintained by developed method in lyophilised form. Process parameters such as temperature, agitation, aeration, pH, pressure, feeding profiles were monitored and controlled during the pilot plant experiment.
