Biochemistry II

Metabolic pathways . Bioenergetics and oxidative metabolism. Short-term and long-term control mechanisms - regulation of enzyme activity, gene expression regulation; cell signaling. Carbohydrate metabolism - major metabolic pathways and their control, special pathways. Tricarboxylic acid cycle. Electron transport chain. Oxidative phosphorylation. Lipid metabolism - utilization and storage of energy. Metabolismus of nitrogen compounds I - Amino acid metabolism. Metabolsim of nitrogen compounds II - Porfyrin and heme metabolism, Nucelotide metabolism. Metabolic interrelationships and control. Compartmentation. Membrane transport. Biochemistry of hormones and the hormonal cascade system.

Learning outcomes of the course unit

The course Biochemistry II is focused on detailed study of metabolic pathways and their regulation on molecular as well as cellular level. The knowlege obtained after passing the course can serve as basis for further studies, especially for biotechnology, molecular biology and bioengineering.

Planned learning activities and teaching methods

The course uses teaching methods in form of Lecture - 2 teaching hours per week. The e-learning system (LMS Moodle) is available to teachers and students.

Examination - oral; discussion of 3 questions form the whole problematics..

Course curriculum

1. Basic principles of metabolism - overview.
2. Metabolic control mechanisms I - regulation of enzyme activity, cell signaling.
3. Metabolic control mechanisms I I - regulation of gene expression, protein turnover.
4. Carbohydrate metabolism - glycolysis, fermentation.
5. Metabolsim of fructose and galactose. Glycogene metabolism and its control. Hormones.
6. Tricarboxylic ycid cycle. Glyoxalate cycle. Glucoenogenesis.
7. Metabolism of lipids and fatty acids. The ketone bodies.
8. Biological oxidations. Electron transport chain and oxidative phosphorylation. Photosynthesis.
9. Isoprenoid pathway. Cholesterol metabolis.
10. Nitrogen metabolism I - fixation, metabolism of amino acids, the urea cycle.
11. Nitrogen metabolism II - porphyrins, heme metabolism. Nucleotides metabolism.
12. Secondary metabolism - overview.

Aims

The aim of the course Biochemistry II is to introduce basic principles of cell metabolism and its regulation. Based on general thermodynamic principles the course of central biochemical pathways including metabolism of saccharides, fatty acids and amino acids is elucidated. Further, basic principles of biological oxidations, cell energy production and photosynthesis are discussed. Next, metabolism of nucleic acids and proteins and processes involved in gene expression are introduced. As a part of course intermediate metabolism and complex regulation of metabolic pathways are presented.
