Methods for Analysis of Biological Systems

The aim of the course is to present methods which are used in chemistry, biochemistry and biotechnology in order to separate, characterize, identify and quantify various biomolecules. The presented methods includes centrifugation, dialysis and ultrafiltration. Furthermore, the course will be focused also on chromatography (gel filtration, ion-exchange chromatography, HIC, affinity chromatography) and its utilization for isolation and purification of proteins and other biomolecules. Moreover, attention will be paid also to electrophoretic techniques used for characterization of proteins (PAGE, PAGE-SDS, Tricine-PAGE, native PAGE, 2D electrophoresis etc.) and DNA/RNA (horizontal electrophoresis, DGGE, TGGE etc.). The aim of the course will be also to explain principles of other methods for characterization of molecular mass of biopolymers such as SEC/MALS, ultracentrifugation, MALDI-TOF, DLS and others. In order to understand structure and function of biomolecules, the students will be familiarized with UV-VIS spectrometry, fluorescence spectroscopy, mass spectrometry, microfluidic techniques, flow cytometry and antibody-based assays . The course will also deals with application of optical, fluorescence and electron microscopy to study complex biological systems.

The students attending the course will gain knowledge as well as practice with selected methods and techniques utilized for analysis of complex biological systems.

Planned learning activities and teaching methods

The course uses teaching methods in form of Lecture - 2 teaching hours per week, Seminar - 2 teaching hours per week. The e-learning system (LMS Moodle) is available to teachers and students.

Examination - written and oral part. Elaboration and prezentation of seminar project.

1. Introduction – definition of biological systems, variability of biological systems
2. Single cell analysis - optical, fluorescent and electron microscopy
3. Single cell analysis – flow cytometry
4. Spectrophotometry, utilization of enzymes as analytical agents
5. Analytical use of luminescent methods in biological sciences
6. Introduction to separation methods – centrifugation, ultrafiltration, precipitation, extraction, dialysis
7. Chromatography for analysis of biomolecules
8. Isolation and purification of proteins
9. Electromigration methods for analysis of proteins and nucleic acids
10. Mass spectroscopy and its use for protein analysis
11. Imunochemistry and its analytical use
12. Selected methods of molecular biology

The aim of the course is to familiarize students with instrumental methods utilized for separation and analysis of biological systems, special attention will be paid to methods used in nanototechnology and nanobiotechnology.
