Originální abstrakt

The aim of this work was study chemical composition and structure of different soil humic acids (HAs). Object of our study were four samples HAs which were isolated from sandy soil – Arenosols (locality Ratíškovice, Czech Republic), Chernozem – Haplic Luvisol (locality Praha-Ruzyně, Crop Research Institute, Czech Republic), brown soil – Gleyic Luvisol (locality Lesonice, Czech Republic) and standard HA Elliott Soil (1S102H). Isolation of soil HAs was performed according to the procedure recommended by the International Humic Substances Society (IHSS). All samples of soil HAs were characterized by elemental analysis, ultraviolet-visible spectroscopy, Fourier transform infrared spectroscopy and steady-state fluorescence spectroscopy. Absorption coefficients (EET/EBz, E4/E6 and delta log K) of soil HAs were calculated from the absorbance values. Infrared spectroscopy is a useful technique in characterization of structure, functional groups and formation modes of HAs. For the fluorescence experiments the final concentration of the HAs was adjusted to 10 mgdm–3. The pH-value of the samples was adjusted to seven using a standard phosphate buffer. Fluorescence mono-dimensional spectra and total luminescence spectra (TLS) of soil HAs were obtained using steady-state fluorescence spectroscopy. All fluorescence spectra were performed on a Horiba Scientific Fluorolog. Fluorescence index (Milori index) of HAs was calculated from the area of the emission spectra. The fluorescence intensity (IF) values (in CPS/MicroAmplitude) of samples were corrected using method of Lakowicz.
