Fluorescent nanoparticles are widely used in various studies as chemically specific markers or as a means of studying the fate of nanosystems in different systems. Furthermore, they can be treated as inert sensors, which by their thermal movement stress and deform their surroundings. By monitoring this movement and correctly interpreting, we can get information about the mechanical properties. In this contribution the influence of environment on particle diffusion studied by fluorescence correlation spectroscopy is discussed. An interesting approach is to convert the time correlations of intensity fluctuations from the FCS method to the time dependence of the particle’s position deviation – a mean square displacement as function of the time. This dependence can be used in microrheological experiments.
