Human transcription factors play critical roles in regulating gene expression and maintaining cellular functions. Understanding their activity and regulation is essential for insights into various biological processes and disease mechanisms. In this study, we investigated the influence of G-quadruplex (G4) prone sequences on the activity of several human transcription factors, including p53, p63, p73, NFkB, and IFI16, using a yeast-based functional assay. Our results show that the presence of a G4 prone sequence adjacent to the target sequences of these transcription factors, derived from their respective target genes, leads to significant changes in their relative activity. This highlights the role of structural DNA features as modifiers of transcription factor functions at target promoter sites. Interestingly, the transactivation in vivo correlated more with the relative propensity of a target site to form cruciforms than with its expected in vitro DNA binding affinity. Therefore, structural features of target sites are significant factors in their transactivation functions.
